چکیده

In vitro dose-response curves are used to describe the relation between chromosome aberrations and radiation dose for human lymphocytes. The lymphocytes are exposed to low-LET radiation, and the resulting dicentric chromosome aberrations follow the Poisson distribution. The expected yield depends on both the magnitude and the temporal distribution of the dose. A general dose-response model that describes this relation has been presented by Kellerer and Rossi (1972, Current Topics on Radiation Research Quarterly 8, 85-158; 1978, Radiation Research 75, 471-488) using the theory of dual radiation action. Two special cases of practical interest are split-dose and continuous exposure experiments, and the resulting dose-time-response models are intrinsically nonlinear in the parameters. A general-purpose maximum likelihood estimation procedure is described, and estimation for the nonlinear models is illustrated with numerical examples from both experimental designs. Poisson regression analysis is used for estimation, hypothesis testing, and regression diagnostics. Results are discussed in the context of exposure assessment procedures for both acute and chronic human radiation exposure.

Benchmark Dose Estimation Using SAS®

Toxicologically-based quantitative risk assessment is concerned with estimating human risks based upon experimental data linking an environmental agent to a known outcome (tumor incidence, acute toxicity, etc.). For dichotomous outcomes dose-response curves are modeled as complex functions of dose which often require specialized software to estimate. The SAS procedure NLMIXED readily allows for...

The growing use of ionizing radiation in various sectors of industry, medicine, agriculture and research has increased the risk of radiation exposure of staff and people. Ionizing radiation as a strong clastogen can cause different types of DNA damages and chromosomal aberrations. Biological dosimetry, based on the cytogenetic analysis of peripheral blood lymphocytes has a wide application in d...

A comparison of algorithms for maximum likelihood estimation of Spatial GLM models

In spatial generalized linear mixed models, spatial correlation is assumed by adding normal latent variables to the model. In these models because of the non-Gaussian spatial response and the presence of latent variables the likelihood function cannot usually be given in a closed form, thus the maximum likelihood approach is very challenging. The main purpose of this paper is to introduce two n...

Retrospective assessment of radiation dose by Fluorescence In Situ Hybridization and evaluation of stable chromosomal aberrations

Estimation of absorbed dose for radiation workers or person involved in different radiological accident is the aim of biodosimetry. Cytogenetic methods are the most current and applicable biodosimetry tools. In chronic or protracted exposure, fluorescence in situ hybridization (FISH), stable chromosomal aberration is used for estimation of absorbed dose. For precise estimation of absorbed dose,...

Maximum Likelihood Estimation of Parameters in Generalized Functional Linear Model

Sometimes, in practice, data are a function of another variable, which is called functional data. If the scalar response variable is categorical or discrete, and the covariates are functional, then a generalized functional linear model is used to analyze this type of data. In this paper, a truncated generalized functional linear model is studied and a maximum likelihood approach is used to esti...
